Bought Full Throttle for my wife's computer (got it on eBay). Thing is: My wife's new machine runs Vista, and from what I understand it won't run games full-screen. I've got DOSBOX, which ran the FT demo well enough (though without any sound or music), though in a small window. Really enjoyed the game when I first played it years back on my ancient Windows 3.11/DOS system, and would like Sherri to enjoy it in the same manner.
So .... how many DOS game fans out there can lend a brother a hand on the best way to run this puppy on my wife's computer. A backup plan is to install the game on our desktop, which runs on XP.

SCUMM games don't play well with Windows XP and Vista.
That's where ScummVM enters the picture. It's bascially a fan made updated SCUMM engine made to run on multiple platforms, even Windows.
I haven't used it myself, but Full Throttle is mentioned as having no significant issues, so it should be able to run it. Try downloading the Windows version, it's an Open Source program. And it will run in full screen, with optional filters, if that's what you want.

While this ScummVM thing does absolutely sound like something you might want to try, I got MI1 and 2 to run flawlessly under XP Pro in fullscreen mode using DosBox.
DFend won't run games properly full-screen (as in has a nasty habit on running them in a window the size of the games' resolution and painting the rest of the screen black when you tell it to run fullscreen), but for DosBox itself it was simply a matter of hitting ALT-ENTER to make the window fullscreen.
In fact I just installed Full Throttle, and it runs in fullscreen just fine. So under XP at least you should be set.
