Fix my error! Please!

Ok, I'm running Vista on this, so I'm thinking I might need to adjust a security setting.
When I run Hijack This!, it starts, then a window pops up and reads, "For some reason your system denied access to the Host file. Hijack This may NOT be able to fix this."
Is there a setting I can change so that this won't happen? I've noticed similar problems when I try to use online virus scanners.
Here's what my clipboard says:

Error #75 - Path/File access error
