Was Grievous killed by the first shot?

I'm of the opinion that all the medical care in the universe couldn't have saved him after one shot. Not even if Palpatine and all his men were right there waiting. There's a bit of a difference between a super-powerful Force user failing to die of shock after sustaining massive trauma, and a non-Force being with his HEART AND LUNGS ON FIRE.

I suppose that depends on what you think the first shot did to Greivous.

It did do SOME burning (We did see flames), but the actual incineration didn't take place until all five shots were fired. Given the unsuaul circumstances and the results, it seems unlikely that teh blaster bolts (directly) struck the gut-sack (we're talking AT LEAST megajoules of energy per shot to incinerate his organs and brain - that much enegy delivered directly into the gut sack would quite easily have exploded it due to vaporization of the liquid content in said organs..)

As I've said before, what probably happened is that the blaster fire struck the interior armor plates of Grevious' chest cavity. Given their superconducting nature, they probably absorbed and "spread out" the energies.. and because of the location of the hits and the arrangement... the energy was radiated INWARDS.. towards his gut sack. In other words, Grievous' chest cavity, when struck by the blaster bolts, turned said cavity into a cremation furnace... with the resulting consequence of cremating his organs (the heat conducting up his neck/body stalk would also cause his brain to explosively vaporize like we saw.)

Given that, it doesnt seem likely that the first shot would have been fatal, though it might have done some harm (his organs would still be getting cooked to some degree by that single bolt.. whether that would be fatal I dont know.)

Yes, particularily since Grievou's brain was vaporized and his vital organs reduced to ashes. Anakin, while severely burned, was not in any way noticably cremated. And comparatively speaking, cremation is a far nastier (and more energy-intensive) feat than even fourth/fifth degree burns.
