I agree. Since the Rebellion was a military organization they should have made background checks on both Luke and Leia. They could after all be imperial spies.
I don't see how DNA evidence would prove or disprove if Luke or Leia themselves were Imperial spies.
There was never any reason to make such a check, so it's perfectly reasonable for them to be ignorant of their relation.
There are many reasons for making such a check. If Luke was killed or was captured how would the Rebellion veify it without DNA evidence ?
How do you verify if someone is captured with DNA evidence when the person ISN'T THERE?
As for if he was killed, there are tons of methods to identify a body. Photos, dental records, fingerprints, blood type, etc... Many of these could result in a positive match without having to resort to DNA evidence.
Besides, how the hell does confirming a corpse's identity result in it being matched and identified as being a family relative, unless they decided to do such a search, which they wouldn't?

He's just making a joke due to the fact that Luke's a farmboy and comparing it to the stereotype (that really more fits with the Apalacian South than with Midwestern farmers, get your regions right, people...) of incestuous relations between family members of the lowest rungs of the class ladder.
The only way the RA would find out that Luke and Leia were blood relatives would be if they decided to see if they matched in the first place, which isn't likely because it would be a waste of time. You don't see it as being a waste of time because YOU already know that they're related, which is clouding your reasoning.
