From the alt.sex.cthulhu FAQ:
HOW DO YOU PRONOUNCE CTHULHU?
1) However he(?) wants you to.
2) Don't pronounce it at all, it only attracts their attention, unless
you want that attention...
3) "...and heard the ominous syllables which can be rendered only as
Cthulhu" - H.P. Lovecraft, Call of Cthulhu.

Or, if you don't want to be obtuse about it:

I hear it pronounced as "Kuh-thool-hoo".
Note: I'm semi-retired from the board, so if you need something, please be patient.

I usually pronounce it "ki-thoo'-loo". It's the way they say it in the movie "Dagon." It's also wrong. That pronounciation can adequately be spelled "cthulu", which it shouldn't be.

The PROPER pronounciation is to almost simultaneously say the C and TH without any vowel between. its quite simple, go to say a hard C (or K) but also position your tongue to say a TH. Say the C then while youre still doing that say the TH then proceed with the ulhu.

From Mr. Lovecraft himself:

"The actual sound - as nearly as human organs could imitate it or human letters record it - may be taken as something like Khlul'-hloo, with the first syllable pronounced gutturally and very thickly."

"The best approximation one can make is to grunt, bark, or cough the imperfectly formed syllables Cluh-Luh with the tip of the tongue firmly affixed to the roof of the mouth. That is, if one is a human being. Directions for other entities are naturally different."
"Heaven is an American salary, a Chinese cook, an English house, and a Japanese wife. Hell is defined as having a Chinese salary, an English cook, a Japanese apartment, and an American wife." -- James H. Kabbler III.
