Load the bed of my truck up with a case of motor oil, gas cans aplenty, a bottle of Tequilla, a shitload of water and canned food, and some camping gear, and drive to Butt-fuck, Wyoming. Gas up before the plague hits the town, then offroad it to a secluded valley, camp for two weeks. Go to San Fransisco, do an "Earth Abides" kinda thing.

That's easy to defend against. No need to make the house airtight, just the basement and ground level windows.

Then go to the grocery store, max out my credit card getting bottled water and canned food, go to the hardware store, get a generator and a shit load of propane, get my friends and family to do the same, and hold up in a suitable house.

Unless you have people counging into your windows, there's no need to even do that. The only one who gets remotely close enough to my house to be a threat would be the mailman. there's no bacterium or virus going to make it through the cracks in your windows in enough amounts to gain a foothold in your body. Extreme measures like driving to Wyoming won't be necessary. Oh, I suppose just to be safe, you could use masking tape. But from outside, the wind currents would swirl it around so much as to cause it to dissipate rather quickly. easy to ride this one out, methinks. Then, when I re emerge, I again emphasize the "Breathes in the oddly fresh air" point again.

Keep in mind, to the rest of the world, its business as normal, the governments are surpressing it because they do not want a riot on their hands.

So, what do you do for the next 48 hours?
The premise of the OP is fundamentally flawed. Any plague that effective will rapidly burn itself out. Furthermore, it would be in a government's best interest to announce that such a monster is about. News like that tends to discourage people from leaving home. (SARS, with it's astonishing 10% lethality rate is a prime example.)
