If you had a majority of the population agree by referendum, had a "republican" ie; no king,form of goverment and were not dead poor ,yes atleast in theory. No new states have joined since the 60's but puerto rico has had a few try's at referendum. There's a secessionist movement in Hawaii, spearheaded primarily by natives. I don't know how powerful it is, though.
